Glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic peptide (GIP) are both incretins - a type of gastrointestinal hormone that causes an increase in the amount of insulin released from the beta cells of the islets of Langerhans after eating, even before blood glucose levels become elevated. GIP and GLP-1 are both secreted within minutes of nutrient ingestion and facilitate the rapid disposal of ingested nutrients. Consequently, in addition to a direct action, this effect of GIP and GLP-1 indirectly reduces insulin release by slowing the rate of absorption of nutrients into the blood stream. As well as inhibiting insulin release, these incretins also inhibit glucagon release from the alpha cells of the Islets of Langerhans.
Both GLP-1 and GIP have been the subject of intense research for the control of diabetes. Since both GLP-1 and GIP are rapidly inactivated by the enzyme dipeptidyl peptidase 4 (DPP-4), one approach is to inhibit DPP-4. Several DPP-4 inhibitors that can be taken orally as a tablet have been developed. For example, Januvia (sitagliptin) was approved by the FDA in 2006.
Stable anaolgs of GLP-1 (7-36) amide have also been developed for diabetes treatment. The main disadvantage of these GLP-1 analogs is that they must be administered by subcutaneous injection.
GIP and GLP-1 secretion remain the focus of research efforts. ReproCELL has developed methods to isolate primary cells which stably secrete GIP and GLP-1 respectively. These cell lines are valuable for the development of assays to identify new therapies for diabetes.
In our assay service, we screen your compound(s) for ability to cause secretion of GLP-1 or GIP from primary rat cells. This is an ELISA based assay in which the cells are typically exposed to the test compound at a range of concentrations for an hour. The cells are not lysed, but the assay media is taken and tested for the presence of the appropriate petide by ELISA.
